This protein is a receptor for urokinase plasminogen activator. It is bound to the plasma membrane via a GPI anchor. However, a soluble protein is also produced in some cell types. It plays a role in localizing and promoting plasmin formation by binding both the proprotein and mature forms of urokinase plasminogen activator (U-PA) and permits the activation of the receptor-bound pro-enzyme by plasmin. The protein is subject to negative-feedback regulation by U-PA which cleaves it into an inactive form.

The mature protein of human UPAR, residues 23 - 335 was over-expressed in HEK293EBNA1 cells and purified to homogeneity. The protein has a C-terminal pre-peptide, residues 306-335, which are removed in the mature protein, 5 potential N-linked glycosylation sites, and 1 GPI-anchor amidated glycine residue. The protein contains a N-terminal hexahistidine tag. The calculated molecular weight without glycosylation of recombinant human UPAR, residues 23 - 335 including hexahistidine tag, is 36.1 kDa. Each vial contains 100 µg UPAR at 0.23 mg/ml.
